If you use WiFi, then no. If you use data, then it will use your data bucket (if you have one) or you'll be charged pay-per-use.

That all depends how long you are using Skype. Honestly, don't use mobile data for Skype at all.

If it's voice calling, it'll be dependant on the quality of the audio. High bitrate = more mobile data used. The average is probably 128kb/sec.

If you do video calling, the usage will be much higher. Again, depending on quality, you would probably be looking at 500kb/sec minimum.

Short bursts of skyping is fine (IE: 5 minute call) as it won't use that much data (IE: % minute call = 37.5 mb if quality is 128kb/sec). Even then, I wouldn't use skype all that much unless it was over wifi, but the choice is yours.
